How To Prepare 50X TAE Buffer (Tris-Acetate EDTA Buffer)

50X TAE Buffer (Tris-Acetate EDTA buffer) Recipe:

1. 1. Tris-base (MW = 121.14 g/mol)        = 242g
   2. 100 % glacial acid                                = 57.1ml
   3. 0.5 M EDTA (pH 8.0)                           = 100ml

Add Enough distilled Water to Prepare the 1 Litter Solution.

Procedure:

1. Weigh 242g Tris-base and dissolve in 700 ml of deionized water.
2. Add Carefully 57.1 ml of 100 % glacial acetic and 100 ml of 0.5M EDTA (pH 8.0)

Add Enough distilled Water to Prepare the 1 Litter Solution.
Note: The pH of this buffer is not adjusted and should be about 8.5

Storage:
Store stock solution at room temperature
